Job description

Adoptive therapy with T cell receptor (TCR)-engineered T cells has demonstrated objective responses in multiple types of tumors, including hard-to-treat solid tumors. To further advance adoptive T cell therapy, it is imperative to counteract deficits in immune control that give way to tumor recurrence. Recurrence of tumor cells is considered a consequence of shortcomings in local CD8 T cell immunity, which in turn is governed by co-stimulatory receptors, such as CD28. In our laboratory, we have designed and tested the therapeutic potential of TCRs equipped with co-stimulatory molecules. These new TCRs provide T cells with the unique ability to activate distinct immune activation and transcriptional programs and demonstrate significant delays in tumor recurrence in immune-competent, tumor-bearing mice.

In the current project, we will study changes in T cell signaling, transcriptomics, energetics as well as immune micro-environments as a consequence of these co-stimulatory TCRs using in silico, in vitro and in vivo models and tools. Aims of this project include: (1) definition of key parameters downstream of T cell co-stimulation that determine effective anti-tumor T cell responses; (2) selection and testing combination therapy with co-stimulatory TCRs; and (3) validation of therapeutic value of co-stimulatory TCRs towards human cancers.

Requirements

You have obtained your PhD in the field of Immunology, preferably T Cell Immunology, you are familiar with preclinical T cell and TCR-based studies. You are able to coordinate and supervise research tasks, able to acquire research funds, and are highly motivated to perform research, take initiatives, communicate easily and cooperate in a team of researchers.

Being able to present a certificate of good conduct, a valid proof of identity, diploma's and/ or relevant registration such as BIG/RGS are conditions for the appointment.

Conditions of employment

We offer a temporary position for 3 years. The gross monthly salary amounts to a maximum of € 4.481,- (scale 10), depending on your level of education and relevant experience and based on a full-time working week of 36 hours. The terms of employment are in accordance with the Collective Bargaining Agreement for University Medical Centers (CAO UMC).
