Job description

Germline mutations in the transcription factor GATA2 result in bone marrow failure with a high predispostion for leukemia. We have modeled this disease in zebrafish by mutating Gata2b, affecting both embryonic and adult hematopoiesis. To be able to establish future gene therapy treatment options for these patients, we need to understand if adult restoration of the Gata2 deficiency is sufficient to abrogate disease ontogeny. For this we have generated a mutant that, upon CRE activation, reexpresses Gata2b from its endogenous locus. The goal of this project is to investigate if reexpression of Gata2b restores the adult leukemia phenotype. The project involves embryonic and adult zebrafish manipulation, flowcytometry analysis and state-of-the-art molecular analyses.

Requirements

The perfect candidate holds a PhD in the life sciences and has experience in zebrafish embryonic development and micromanipulation and experience using flowcytometry. Applicants with a background in (malignant) hematopoiesis and embryonic development are also encouraged to apply.

Conditions of employment

  • The gross monthly salary amounts a maximum of € 4.800,- (scale 10), depending on your level of education and relevant experience, based on a full-time working week of 36 hours.

The research will be conducted within the de Pater group within the department of Hematology, supported by two technicians. The department of hematology is an international department and we host national and international speakers for our Hematology lecture series to improve collaborations. As we are both a basic and clinical department, we have access to both state-of-the-art sequencing and imaging equipent required for this project and patient samples to translate the findings of this project to clinical care.
