Job description

Nucleotide Excision Repair (NER) is a versatile DNA repair pathway that preserves genomic integrity by removing different types of DNA-helix distorting lesions. Hereditary defects in NER are associated with distinct, clinically heterogeneous diseases characterized by developmental failure, neurological defects, cancer predisposition and segmental progeria. In patients, NER deficiency affects tissues differently, which is currently not understood. In this project, we aim to develop new human in vitro differentiated cellular models to study the causes of this tissue-specificity at the molecular level, to elucidate the exact pathogenesis of NER disease. As NER protects against cancer and aging, studying NER function in different cell types is crucial to better understand human health.

The PhD candidate will participate in this novel and exciting line of research, focusing on the development of new in vitro cellular differentiation models, based on iPS and CRISPR-Cas9 technology, to dissect NER activity and organization, as well as the impact of patient-derived NER mutations, specifically in differentiated cell types. To this end, the PhD candidate will employ a multidisciplinary approach, using various DNA repair assays, genetics, live confocal imaging, quantitative proteomics and transcriptomics.

Requirements

We are looking for a highly motivated and talented candidate that holds a MSc degree in Biomedical Science, Molecular/ Cellular Biology, Biochemistry or related field with a basic knowledge of DNA repair pathways. Experience with standard molecular biology techniques, live cell imaging and proteomic tools will be a strong advantage. Excellent writing and presentation skills in English are recommended. As you will work in a dynamic international research environment we need a team-player who is able to combine independent thinking with a collaborative attitude.

Being able to present a certificate of good conduct, a valid proof of identity, diploma's and/ or relevant registration such as BIG/ RGS are conditions for the appointment.

Conditions of employment

You will receive a temporary position for 4 years. The gross monthly salary is € 2.279 ,- in the 1st year and increases to € 2.919,- in the 4th year (scale OIO). The terms of employment are according to the Collective Bargaining Agreement for Dutch University Medical Centers (CAO UMC).
