Job description

Genomes are subject to continuous damaging insults from both environmental and endogenous sources such as the cellular metabolism. Unrepaired DNA lesions can impede both DNA and RNA polymerases, resulting in replication stress and transcription stalling. We have recently discovered that genome-wide transcription stalling is present in metazoan aging with striking similarities to DNA damage accumulation using a combination of newly developed sequencing methods (manuscript submitted). In this exciting and challenging project, the postdoctoral researcher will further unravel the mechanisms of age-related transcription stalling by an integrative genomics/transcriptomics approach using state-of-the-art transcriptomics and genomics datasets, including de novo RNA synthesis sequencing, RNA polymerase II ChIP-sequencing, single cell RNA sequencing, nanopore sequencing and proteomics. These studies will provide the mechanistic framework of transcription stress in aging. Studies will be performed in close collaboration with experimental biologists.

Requirements

The highly motivated and successful candidate must have a PhD degree in a relevant discipline (Bioinformatics, Computational Biology, Statistics, Computer Science) and proficient in "R" and/or other languages such as Python. The candidate should be acquainted with analyzing bulk- and single-cell sequencing data with profound understanding of mathematical and statistical approaches and is capable of generating new mathematical models to address different challenges and scenarios in the biological samples. Experience with machine learning, the basal transcription process, DNA damage responses and/or aging will be considered an advantage. The candidate should be able to independently design, conduct and evaluate research projects, have outstanding theoretical and experimental research skills, is well-organized and result-driven with a strong team spirit. Furthermore, a strong command of (scientific) English in speech and writing is essential, proven by publications in international journals and presentations at international meetings.

Being able to present a certificate of good conduct, a valid proof of identity, diploma's and/ or relevant registration such as BIG/ RGS are conditions for the appointment.

Conditions of employment

The gross monthly salary amounts a maximum of € 4.361,- (scale 10), depending on your level of education and relevant experience and based on a full-time working week of 36 hours. The terms of employment are in accordance with the Collective Bargaining Agreement for University Medical Centers (CAO UMC).
