Lipid metabolism is tightly regulated both at the cellular as well as whole-organism level. This regulation ensures that the level of lipids does not increase to toxic levels and that they are available at sufficient amounts for processes requiring them. Disruption of this balance is at the center of several pathological conditions in humans, most notably fatty liver and cardiovascular disease.

Our group is broadly interested in the transcriptional and post-transcriptional regulation of lipid and energy metabolism. We recently applied genome-wide functional genetic screens to identify novel genes that play a role  in these processes (Loregger et al. ATVB 2017; Tan et al. Atherosclerosis 2019, Loregger et al. Nat. Comm, in press). In this VICI-supported project we will apply CRISPR/Cas9- or haploid-based genome-wide functional genetic approaches to study lipid-associated cellular processes including, amongst others, cholesterol and fatty acid synthesis and intracellular sterol trafficking. The goal of this project is to identify novel genes and elucidate the molecular mechanisms that govern these processes, and investigate whether these play a role in cardiovascular and metabolic disease. A wide range of advanced techniques will be used in this project, including state-of-the-art genetic screens, cell-based functional assays, metabolomics, mass spectrometry, and molecular biology.

We are looking for a broadly-trained and highly-motivated PhD candidate with an MSc in Medical Biology, Life Sciences, Biochemistry or a related field. Experience and/or affinity with functional genetic screens, next-generation sequencing, or bioinformatics is an advantage. The candidate should have good command of oral and written English. Excellent communication skills and the ability to work within an international research group are essential. The appointment will be for 4 years.
