Job description

ERCC1-XPF is an endonuclease complex that has essential functions in multiple DNA repair pathways and is therefore one of the major cellular defence factors protecting against mutation, cancer and aging. The medical importance of ERCC1-XPF is illustrated by the severe cancer-prone and progeroid diseases, such as xeroderma pigmentosum, Cockayne syndrome and Fanconi anemia, that are caused by its deficiency. ERCC1-XPF has been investigated in much detail in relation to its function in nucleotide excision repair, which is a versatile DNA repair pathway that removes DNA damage caused by solar UV light and certain cancer chemotherapeutics. However, ERCC1-XPF function in other DNA repair pathways, including interstrand crosslink repair, is less well investigated.

In this project, the PhD candidate will study cellular mechanisms that control ERCC1-XPF activity in DNA repair and characterize novel DNA repair mechanisms in which ERCC1-XPF is involved. To this end, a multidisciplinary approach will be employed, with molecular, genetics and cell biological techniques ranging from DNA repair assays, genetic screening, CRISPR-Cas9 gene editing to confocal imaging and quantitative proteomics, using both human cells and C. elegans as model systems.

Requirements

  • We are seeking a highly motivated and talented candidate that holds a MSc degree in Biomedical Sciences, Molecular/Cellular Biology, Nanobiology, Biochemistry or a related field.
  • Experience with standard molecular biology techniques and live cell imaging will be a strong advantage.
  • As you will work in a dynamic international research environment, we need a team-player who is able to combine independent thinking with a collaborative attitude.
  • A strong command of (scientific) English in speech and writing is essential (EUR requirements minimum IELTS overall score of 7.0 [comparable to TOEFL PBT: 600; TOEFL CBT: 250; TOEFL IBT: 95]).

  • You will receive a temporary position for 4 years.
  • The gross monthly salary is € 2.789,- in the 1st year and increases gradually to € 3.536,- in the 4th year (scale OIO).

The Department of Molecular Genetics has a long-standing world-wide established reputation in dissecting the DNA damage response at the molecular, cellular and physiological level. The department consists of a vibrant and international group of more than 80 researchers working on various aspects of DNA damage, DNA repair and related pathways. This project will be carried out in the group of Dr. Hannes Lans , which investigates molecular mechanisms of the DNA damage response in relation to DNA repair disease and cancer therapy. For this, we use molecular genetics and cell biological techniques in both mammalian cells and C. elegans and strongly focus on state-of-the-art live cell imaging.
