Job description

Nucleotide Excision Repair (NER) is a versatile and important DNA repair pathway that protects against cancer and aging by removing different types of helix-distorting DNA damage. TFIIH is a dual-function transcription/DNA repair complex that plays a central role in NER, by opening of the DNA helix and verifying the presence of DNA damage, and in transcription, by helping RNA polymerase II initiate RNA synthesis. Intriguingly, hereditary defects in TFIIH are associated with several distinct, clinically heterogeneous diseases characterized by cancer predisposition, progressive neurological defects, developmental failure and segmental progeria, whose pathogenesis is not fully understood. As NER protects against cancer and aging, studying TFIIH function is crucial to better understand human health.

In this project, the PhD candidate will investigate the biological impact, and underlying mechanism, of TFIIH mutations, as well as the role and activity of novel identified proteins that regulate TFIIH and NER function. To this end, a multidisciplinary approach will be employed, with molecular, genetics and cell biological techniques ranging from various DNA repair assays, CRISPR-Cas9 gene editing, and live confocal imaging to quantitative proteomics and transcriptomics. Furthermore, both human cells and C. elegans will be used as model systems.

Requirements

  • We are seeking a highly motivated and talented candidate that holds a MSc degree in Biomedical Science, Molecular/Cellular Biology, Nanobiology, Biochemistry or a related field. Experience with standard molecular biology techniques, live cell imaging and/or C. elegans will be a strong advantage.
  • As you will work in a dynamic international research environment, we need a team-player who is able to combine independent thinking with a collaborative attitude.
  • A strong command of (scientific) English in speech and writing is essential.

Conditions of employment

You will receive a temporary position for 4 years. The gross monthly salary is € 2.570,- in the 1st year and increases to € 3.271,- in the 4th year (scale OIO). The terms of employment are according to the Collective Bargaining Agreement for Dutch University Medical Centers (CAO UMC).

The Department of Molecular Genetics has a long-standing world-wide established reputation in dissecting the DNA damage response at the molecular, cellular and physiological level. The department consists of a vibrant and international group of more than 80 researchers working on various aspects of DNA damage, DNA repair and related pathways.

This project will be carried out in the group of dr. Hannes Lans, which investigates molecular mechanisms of DNA repair, in particular NER, in relation to DNA repair disease and cancer therapy. For this, we use molecular genetics and cell biological techniques in both mammalian cells and C. elegans and strongly focus on state-of-the-art live cell imaging. Our group collaborates intensively with the group of Prof. dr. Wim Vermeulen and together we act internationally at the forefront of the DNA repair field.
