We are seeking an ambitious PhD student to join our multidisciplinary team. Your role will involve investigating the distinctive immune features involved in pediatric T and B cell interactions and establishing a platform to explore these variations.

Job description

The basis of long-lasting protective humoral immunity (antibodies) is initiated upon the intimate interaction between naïve T and B-cells in lymphoid organs. It is widely acknowledged that the immune system of children differs from that of adults, leading to more frequent infections and the need for repeated vaccinations. However, children are still treated as small adults, rather than leveraging these differences to benefit pediatric health.

Recent research, including our own, has shown a distinctive immune signature in naïve T cells that is particularly abundant in young children. This signature suggests that these T cells may respond differently to environmental stimuli. In this four-year funded project, you will investigate how this unique immune profile influences the interaction between naïve T and B cells, B cell functionality, and whether it can counteract the perceived immune immaturity in children. In the end, identified immune stimuli could be utilized as potential age-specific vaccine adjuvants.

To achieve this goal, you will employ a range of in vitro assays, flow cytometric and transcriptomic analysis, and imaging techniques to explore various T:B cell interactions and their impact on B cell behavior. Additionally, you will establish a lymphoid tissue model (3D organoid) to quickly assess different immune stimuli and to enhance the translation of our in vitro findings.

Requirements

We seek a skilled and driven PhD candidate with a background in biomedical sciences. You should possess outstanding experimental abilities and ideally have some experience in cell culture, flow cytometry, ELISA, and analysis of large datasets. Affinity with early-life immunology is desirable, along with a curiosity to investigate various lymphoid models. Effective communication and organizational skills are essential. The ideal candidate will demonstrate flexibility, adaptability, and a strong ability for teamwork.

Conditions of employment

The maximum salary for this position (36 - 36 hours) is € 3.677,00 gross per month based on full-time employment.

In addition, we offer an annual benefit of 8.3%, holiday allowance, travel expenses and career opportunities. The terms of employment are in accordance with the Cao University Medical Centers (UMC).

Department

You will be based in the laboratory of Dr. van den Broek, specializing in experimental immunology, situated within the Center of Translational Immunology (CTI) at UMC Utrecht. Dr. van den Broek's research group is newly established, dedicated to unraveling the complexities of T and B cell interactions in both health and disease, as over time. The group actively engages in collaborative efforts on related projects.

The CTI is a well-equipped department boasting expertise across all aspects of immunology and is integrated into the broader focus area of 'Infection and Immunology' at UMC Utrecht. Our structure is designed to facilitate seamless translation from laboratory findings to clinical applications. Within CTI, or through collaborations with UMCU/UU, access to cutting-edge technologies such as flow cytometry (spectral flow), imaging (confocal, multi-photon), mass spectrometry, and scRNA sequencing is readily available.

As part of this project, you will also participate in group meetings with the van Wijk group, specializing in tissue immunology and T cells, and collaborate with Dr. Heesters' group, which investigates germinal center biology.
